I have 2 Amex cards==Platinum and Blue Cash. I use Blue Cash exclusively for groceries. Both cards transactions show Amazon as a payee but the Blue Cash Amazon charges are for Whole Foods deliveries through Amazon. Platinum charges to Amazon are all Shopping. Is there a way to have Quicken distinguish between the two and assign one category as Shopping and another as Groceries--a formula that I can apply based on which card was used? Or do I have to manually regularly change the transactions' catgory?

Unfortunately there isn't any way to do this in Quicken. Ideally you would be able to change the renaming rule based on the account, and therefor direct it to a different memorized payee, but renaming rules and memorized payees for that matter are global.
